INTRODUCTION
TNF-α is a cytokine produced by monocytes and macrophages. It consists of a 35 amino acid (aa) cytoplasmic domain, a 21 aa transmembrane segment, and a 177 aa extracellular domain. Cleavage of membrane bound TNF-α by TACE/ADAM17 releases a 55 KDa soluble trimer. TNF-α functions as a modulator of immune response and plays a role in the pathogenesis of many disease states.
